Chisa Yukizome, a former student at Hope's Peak Academy gets hired as a new teacher for Class 1 - B. However, most of the students didn't show up for class, leaving Chisa, Fuyuhiko Kuzuryu, Hiyoko Saionji, Mahiru Koizumi, Sonia Nevermind and Mikan Tsumiki to look for them. But things can get pretty crazy when the Ultimates are at play.

Koichi Kizakura recalls his reasons for staying with Kyoko Kirigiri, who works to uncover the truth behind Sonosuke Izayoi's murder. But just being at the crime scene is dangerous enough itself. Meanwhile, Byakuya Togami looks for a way to break the Future Foundation out of the building, but certain obstacles prevent him from reaching that goal.
After noticing that Mikan has been acting strangely around Junko, Ryota is shocked to discover a video of the student council's killing game. Junko reveals that she plans to combine this video with Ryota's anime to create something that will brainwash people and send them into despair, threatening to subject his classmates to another killing game should he refuse to cooperate.
Betrayals are committed as Ruruka Ando struggles to survive and the traitor is close to being revealed. Monaca Towa's prophecy comes true when Kyoko Kirigiri's NG code is violated, because of Makoto Naegi. To bring an end to this dreadful game of despair, Kyosuke Munakata decides it's time to decide whose hope is better: his or Makoto's.
Chisa shows up to confront Junko, who reveals her plan to brainwash her class into Ultimate Despairs, showing its effectiveness by having a student kill himself on command. Meanwhile, Chiaki escapes with Nagito and informs the rest of the class, who decide to go help their teacher.
Following Kyoko Kirigiri's death, Makoto Naegi resolves to bring an end to the Final Killing Game and clashes with Kyosuke Munakata, fighting for whose hope is stronger. However, a recent, shocking revelation about Chisa Yukizome has driven Kyosuke into insanity and despai
Junko overpowers Juzo and deduces that he is in love with Kyosuke, threatening to expose his feelings unless he betrays Kyosuke by reporting that she is innocent. Meanwhile, Chisa, who has been brainwashed by Junko's despair video, sends Chiaki down to an underground room where she is captured by Junko. Deciding that Chiaki is the final element needed to finish her prototype, Junko forces her to go through a dungeon filled with deadly traps, while the rest of the class are forced to watch.
Leaving behind her notebook, Kyoko Kirigiri reveals the truth behind the Final Killing Game, leaving Makoto Naegi to test out his late friend's theory, only to be shocked by what he sees. Meanwhile, Juzo Sakakura decides to bring an end to this dreaded game in order to save Kyosuke Munakata.
The blackmailed Juzo and the brainwashed Chisa both tell Kyosuke that Junko is innocent, allowing her plan to continue unhindered. As the 77th class graduate from their classroom to spread despair across the world, the Reserve Course students storm the main building and kill everyone inside before Junko uses her despair video to have them all commit suicide.
Kazuo Tengan's motives for masterminding the Final Killing Game are revealed as Ryota Mitarai decides to put an end to despair, by brainwashing all of humanity to sacrifice all negative emotions and unconditionally believe in hope. Makoto Naegi, Aoi Asahina and Kyosuke Munakata try to stop him, but their efforts are impeded by the brainwashed Future Foundation.
As Makoto races to stop Ryota from broadcasting his Hope video, he and the others are assisted by the arrival of the 77th Class, who Hajime managed to restore to normal using Izuru's talents. After making their way past a hypnotised task force, Hajime and the others confront Ryota, who blames himself for the entire incident. Feeling that he should not have to carry his burden alone, Hajime and the others ask Ryota to come with them as they aim to atone for their sins, convincing him to cancel the broadcast.
